Reconstructing promoter activity from Lux bioluminescent reporters
The bacterial Lux system is used as a gene expression reporter. It is fast, sensitive and non-destructive, enabling high frequency measurements. Originally developed for bacterial cells, it has also been adapted for eukaryotic cells, and can be used for whole cell biosensors, or in real time with live animals without the need for euthanasia. However, correct interpretation of bioluminescent dat...
متن کاملOptimisation of Bioluminescent Reporters for Use with Mycobacteria
BACKGROUND Mycobacterium tuberculosis, the causative agent of tuberculosis, still represents a major public health threat in many countries. Bioluminescence, the production of light by luciferase-catalyzed reactions, is a versatile reporter technology with multiple applications both in vitro and in vivo. In vivo bioluminescence imaging (BLI) represents one of its most outstanding uses by allowi...
متن کاملMultiplexing Bioluminescent and Fluorescent Reporters to Monitor Live Cells
Reporter proteins are valuable tools to monitor promoter activities and characterize signal transduction pathways. Many of the currently available promoter reporters have drawbacks that compromise their performance. Enzyme-based reporter systems using cytosolic luciferases are highly sensitive, but require a cell lysis step that prevents their use in long-term monitoring. By contrast, secreted ...
متن کاملObelin mutants as reporters in bioluminescent dual-analyte binding assay
Two obelin mutants distinctly different in the color of bioluminescence were successfully applied to simultaneous detection of two analytes in a single well. The flash-type signals were triggered by single injection of Ca and were discriminated by the spectral and time resolutions. The technique was developed in a high-throughput format, and applied to simultaneous immunoassay of two gonadotrop...
متن کاملConstruction of mobilizable mini-Tn7 vectors for bioluminescent detection of gram-negative bacteria and single-copy promoter lux reporter analysis.
We describe the construction of mini-Tn7-based broad-host-range vectors encoding lux genes as bioluminescent reporters. These constructs can be mobilized into the desired host(s) by conjugation for chromosomal mini-Tn7-lux integration and are useful for localization of bacteria during infections or for characterizing regulation of promoters of interest in Gram-negative bacteria.
